There are a number of possibilities for why this is occurring. First, make sure the iSQL*Plus service is running. For the Windows operating system, there should be a service called "Oracle<OracleHomeName>HTTPServer" in the Services panel. Start this service and then try to launch the iSQL*Plus URL again.
If unsuccessful, ensure that the <hostname.domainname> provided in the URL can be resolved. Use the IP address instead of the <hostname.domainname> in the URL to see whether that resolves the problem.
